Remarks Nowadays the demands for most sensitive animal species detection in particular of pork is becoming of increasing interest. The molecular biological real-time PCR method, based on specific DNA amplification detects highly specific DNA and DNA fragments of the relevant species. The sensitivity is based on the detection system and the food matrix and processing grade. Determining Ct values of the real-time PCR run, relative quantification is possible. However systems are calibrated on raw muscle tissue. In this ring trial stable gelatin is used as a matrix for qualitative detection of low amounts of present porcine DNA. Based on the former successful profiency test RVEP 150204 whereas porcine DNA (fragments) were specifically identified in gelatin for human consumption, the new profiency test will include the sensitive detection of pork (sus scrofa) and optionally also of bovine (bos taurus). The PT is designed to use DNA methods (preferentially qPCR), but other tests might be used as well. The profiency test will consist of a sensitive qualitative detection of traces of DNA (fragments). Even so the achieved ct values will indicate how strong the results and therefore sensitivity of different systems are. A real quantification will not be possible in this matrix.
